The intensity of the input from one eye is different, so that the degree of pupillary constriction with a light shining in one pupil is less than when with the light shining in the other pupil. Common causes include optic neuritis (Multiple Sclerosis - MS), and retinal lesions. Lesions of the optic pathways that are distal to the lateral geniculate body will NOT produce an afferent pupillary defect.
